Biological repository of cells and supernatant of induced sputum, saliva, buccal cells.

Induced sputum specimens.

Induced sputum specimens have been collected since 2008. Specimens of induced sputum cells and supernatant from 217 individuals (147 males and 70 females) are stored in the freezers at–75°C – 80°Ñ.

Radiochemical and plutonium workers exposed to internal (alfa-) and external (gamma-) radiation – 141 individuals (114 males, 27 females), auxiliary plants workers – 13 individuals.

Pu body burden for the examined individuals ranges from 0.04 to 6.64 kBq, average absorbed dose of gamma-radiation from Pu to lung was from 0.01 to 1.38 Gy. Absorbed external gamma dose was from 0.003 to 1.67 Gy for the main group during their occupational career.

Average age of the examined workers was 70.7 ± 0.5 (45 ? 85), of the control group – 64.7 ± 1.2 (40 ? 82, n = 67).

Medical data were collected, incidence analysis was conducted, all cases of respiratory diseases were ascertained, and diseases were classified in compliance with ICD-9. Number of examined Mayak PA workers with respiratory diseases was 84%. The most common diseases in the incidence structure were chronic bronchitis (48.4%), nasopharyngitis (34.4%), and chronic obstructive pulmonary disease (31.2%). Percentage of smokers among the main plants workers was 32.7%, in the control – 23.3%.

Buccal cells specimens.

As a rule buccal cells for storage were collected at the same time with blood collection. Collection and preparation of cells was done in compliance with the NCI USA “Cell culture” protocol and recommendations for researchers (Harty L.C., Shields P.G et al., 2000). After step-freezing in CryoMed Freezing Chamber (Forma Scientific, USA) 1493 cells specimens were placed on long-term storage in freezer at temperature from –75°Ñ to –80°Ñ in number:

  • (0,1–1,0)Å+6 cells (31%);
  • (0,1–5,0)Å+6 cells (50%);
  • (5,1–10,0)Å+6 cells (15%);
  • > 10,0Å+6 cells (4%).

Specimens of cells and supernatant of buccal cells from 1493 individuals (897 males, 581 females) were collected, treated and stored in the Repository freezers.

Saliva specimens.

Saliva specimens have been collected since 2011. Saliva specimens have been obtained from 103 individuals (64 individuals –Mayak PA main plants workers, 39 individuals – Ozersk residents). Obtained and cooled specimens were transported to the laboratory (309 storage items, 3 vials per each specimen), after treatment frozen for 1 hour at– 20°Ñ, then stored at –70°Ñ. Immediately before the analysis a specimen is defrosted and centrifuged.
